Artesunate: A natural product-based immunomodulator involved in human complement

چکیده

• ART possesses anti-complement bioactivities. is involved in three effector pathways of complement interception. targets C1q likely via binding globular head region. specifically binds to the key therapeutic C1q, C3 and C5. A modified ELISA selectively allows activation. Complement an important innate immune defence machinery. Once dysregulated, it often linked pathogenesis diverse autoimmune diseases. Artesunate (ART) a well-known anti-malarial compound. Recently, has been highlighted by its potential effects on certain complement-related However, underlying mechanisms are hitherto unknown. In present study, we found that mediated interception as validated analysis haemolytic assay. cell-based setup using dying Jurkat cells, ART-mediated was also confirmed. Further, newly established system allowing activation classical pathway, lectin pathway alternative respectively. revealed dose-dependently inhibited C4 activation, terminal complex assembly pathways. blockade C5 with lesser extent properdin. The interaction determined be FACS ART-conjugated mesoporous silica particles bound microparticles. conclusion, for first time report bioactivities suggest benefit human

Natural IgM antibodies are involved in the activation of complement by hypoxic human tubular cells.

Ischemia-reperfusion injury (IRI) has a major impact on graft survival after transplantation. Renal proximal tubular epithelial cells (PTEC) located at the corticomedullary zone are relatively susceptible to IRI and have been identified as one of the main targets of complement activation. Studies in mice have shown an important role for the alternative pathway of complement activation in renal ...

Aminosugar-based immunomodulator lipid A: synthetic approaches

The immediate immune response to infection by Gram-negative bacteria depends on the structure of a lipopolysaccharide (LPS, also known as endotoxin), a complex glycolipid constituting the outer leaflet of the bacterial outer membrane. Recognition of picomolar quantities of pathogenic LPS by the germ-line encoded Toll-like Receptor 4 (TLR4) complex triggers the intracellular pro-inflammatory sig...
